Summary
Within the nucleus of our cells, two meters of genetic material get folded in an organized manner to guarantee compaction and a proper pattern of gene expression. Understanding how this spectacular folding is achieved is key to unravel the bases of gene regulation in health and disease. However, the mechanisms controlling genome folding, in particular at the single gene level, are still largely unknown.
In this context, we focus on investigating how does gene conformation and gene activity influence each other.
To address this question, we integrate advanced Super-resolution imaging strategies (STORM, Single Molecule Tracking, OligoSTORM, DNA-PAINT, PoSTAC) with quantitative analysis to resolve gene organization at the molecular level in single nuclei. Also, we develop ad hoc tools to label endogenous genomic regions allowing the study of gene dynamics at high temporal and spatial resolution (Neguembor et al. Nucleic Acids Res 2018).
